The aim of this study was to evaluate the effect of premarin, a natural compound derived from natural products, on the activity of the CYP3A4 metabolizing enzyme, CYP2D6. We examined the effect of premarin on the CYP2D6 enzyme activity of human plasma.
Background:CYP3A4 inhibitors have been shown to reduce the metabolism of the major drug class of drugs in the human body. Since the enzyme CYP2D6 is associated with inhibition of CYP2D7, it is important that these drugs be used with caution.
Methods:We conducted a prospective study in 826 men with abnormal uterine bleeding who were treated with the combined oral contraceptive implant. All the patients were aged ≥ 30 years. All the patients had a history of a previous cardiovascular disease. Blood samples were obtained before and after the implantation of the implant. Plasma was separated by centrifugation, separated using glass capillaries, and stored at -80 degrees C to be analysed for the presence of CYP2D6. Plasma concentrations of the active CYP2D6 were determined by high-performance liquid chromatography (HPLC). The plasma concentration of active CYP2D6 was also determined. The plasma levels of CYP2D6 metabolizing enzymes (CYP3A4, CYP2D6 and CYP2D6-3A4) were assessed using enzyme-linked immunosorbent assay kits.
Results:The plasma levels of CYP3A4 and CYP2D6-3A4 were significantly higher in the patients treated with premarin compared to those treated with the drug alone. Plasma concentrations of the active CYP2D6 metabolizing enzyme (CYP3A4 and CYP2D6-3A4) were significantly higher in the patients treated with premarin compared to the patients treated with the drug alone. Concomitant treatment with the drug alone did not affect the levels of the active CYP2D6 metabolizing enzyme in these patients. Concomitant treatment with premarin resulted in significantly lower levels of CYP3A4 and CYP2D6-3A4 compared to the drug alone. Concomitant treatment with the drug alone resulted in significantly lower levels of the active CYP2D6 metabolizing enzyme compared to the drug alone. The plasma levels of CYP3A4 and CYP2D6-3A4 were also significantly lower in the patients treated with premarin compared to the patients treated with the drug alone.
Conclusion:In conclusion, premarin is a natural compound derived from natural products and has an effect on the activity of the enzyme CYP2D6 and possibly other enzyme-modifying drugs.

How to use PREMARIN VAGINAL?
Apply PREMARIN VAGINAL as directed by your physician. It should be used only in and around the vagina or the genital area. PREMARIN VAGINAL comes in a tube along with an applicator. Inform your doctor about any unusual vaginal bleeding, as postmenopausal vaginal bleeding could indicate uterine cancer. Share your complete medical history, especially conditions like asthma, epilepsy, diabetes, migraine, endometriosis, lupus, heart, liver, thyroid, kidney issues, or elevated blood calcium levels.
